Intended Use
Typhoid IgG/IgM Antibody Test Kit (Immunochromatographic Assay) adopts the colloidal gold method to qualitatively detect the antibody of typhoid bacillus (lipopolysaccharide antigen and outer membrane protein antigen) in human serum / plasma, which is suitable for the early auxiliary diagnosis of typhoid infection.

Test Principle
Typhoid IgG/IgM Antibody Test Kit (Immunochromatographic Assay) is a lateral flow chromatography immunoassay. The test cassette consists of: 1) a burgundy colored conjugate pad containing recombinant S. typhoid H antigen and O antigen conjugated with colloid gold (Typhoid conjugates) and rabbit IgG ‐gold conjugates, 2) a nitrocellulose membrane strip containing two test bands (IgG and IgM bands) and a control band (C band). The IgM band is pre-coated with monoclonal anti-human IgM for the detection of IgM anti-S. typhi, IgG band is pre-coated with reagents for the detection of IgG anti-S.typhi , and the C band is pre-coated with goat anti rabbit IgG.

Operation Flow

Step 1: Sampling

Collect human Serum/Plasma/Whole blood properly.

Step 2: Testing

1. Remove an extraction tube from the kit and a test box from the film bag by tearing the notch. Open the inspection card aluminum foil bag. Remove the test card and place them horizontally on a table.

2. Use a disposable pipette, transfer 4μL serum (or plasma), or 4μL whole blood into the sample well on the test cassette.

3.Open the buffer tube by twisting off the top. Put 3 drops (about 80 μL) of assay diluent into the assay diluent well round shaped. Start counting.

Step 3: Reading

Read the result at 15 min. Results after 20 minutes are invalid.

Result Interpretation

Negative result
Only the quality control line C appears and the detection lines G and M do not show, it means that no Typhoid antibody is detected and the result is negative.

Positive Result
If both the quality control line C and the detection line M appear= the Typhoid IgM antibody is detected, and the result is positive for the IgM antibody.
If both the quality control line C and the detection line G appear= the Typhoid IgG antibody is detected and the result is positive for the IgG antibody.
If both the quality control line C and the detection lines G and M appear= the Typhoid IgG and IgM antibodies are detected, and the result is positive for both IgG and IgM antibodies.

Invalid Result
If the quality control line C cannot be observed, results will be invalid
regardless of whether a test line shows, and the test should be repeated.
